Effects of the isoflavone puerarin and its glycosides on melanogenesis in B16 melanocytes

چکیده
We investigated the effects of puerarin (8-Cglucosyl-7,40-dihydroxy isoflavone), an isoflavone found in Kudzu roots (Pueraria lobata), and its glycosides (enzymatically synthesised, water-soluble derivatives of puerarin) on melanogenesis in vitro. Puerarin and its glycosides reduced mushroom tyrosinase activity by 88 and 67% at 4.8 mM, respectively, in a concentration-dependent manner. The puerarin glycosides were less effective than puerarin at the same concentration but showed a comparable inhibitory effect at a concentration at which puerarin is insoluble in water. In cultured B16 melanocytes, the melanin content was reduced significantly; moreover, tyrosinase activity was inhibited significantly by both puerarin and its glycosides when added at a concentration of 480 lM. DNA microarray and RT-PCR analyses showed significant downregulation of the expression of microphthalmia-associated transcription factor (MITF) and its target genes. The protein expression of MITF and tyrosinase was also downregulated significantly by 40 and 50%, respectively. Our findings suggest that puerarin and its glycosides cause hypopigmentation via dual mechanisms: by inhibiting tyrosinase activity directly and by altering the expression of melanogenesis-related genes, such as MITF and tyrosinase. Therefore, puerarin and its glycosides may have potential for the development of functional cosmetics causing hypopigmentation.

Antioxidative and hypocholesterolemic activities of water-soluble puerarin glycosides in HepG2 cells and in C57 BL/6J mice.
Puerarin is an isoflavone derived from Kudzu roots and has antioxidant and hypocholesterolemic effects; however, its insolubility often limits its biological availability in vivo. Using a novel transglycosylation process, the solubility of puerarin glycosides was increased >100-fold, but it was not known whether these modified puerarin glycosides maintained biological activities. We found that ...

Method development for determination of isoflavones in kudzu was achieved by HPLC/UV/ESI-MSD. Using three kudzu species of Pueraria lobata, P. thomsonii and P. edulis, and analyzing the isoflavones separately by species and from different plant tissues (roots, stems, leaves, flowers and fruits) in each species, a total of 25 isoflavones were identified by their molecular ions and characteristic...

Objective(s): Nepeta binaludensis Jamzad (Lamiaceae) has been used in folk medicine of Iran to cure various diseases. The plant is an endemic species to the country that has recently been identified in Razavi Khorasan province. To evaluate the antioxidant and anti-melanogenesis of N. binaludensis, in this study the inhibitory activity of different extracts of N. binaludensis in murine melanoma ...

To investigate the new physiological functions of oolong tea, the effects on melanogenesis were studied. An oolong tea extract inhibited melanogenesis without affecting cell growth in B16 mouse melanoma cells. However, the oolong tea extract hardly showed any inhibitory effect on mushroom tyrosinase in a cell-free system. The effects of an oolong tea extract on the intracellular tyrosinase leve...
متن کاملControl of phenotypic expression of cultured B16 melanoma cells by plant glycosides.
The effects of two plant glycosides, ginsenosides Rh1 and Rh2, on the growth and differentiation of mouse melanoma (B16) cells in culture were studied. These plant glycosides have a dammarane skeleton resembling a steroid skeleton as an aglycone. Ginsenoside Rh2 inhibits the growth of B16 melanoma cells, causes morphological alterations, and stimulates melanogenesis at high cellular density. Wh...
